A single cell-based system to investigate mechanisms of neurodegeneration

Resumen: Using a methodology based on automated microscopy and longitudinal survival analysis we study molecular mechanisms of neurodegeneration. Specifically, I will introduce a neuronal model of alpha-synuclein in which we have determined how alpha-synuclein expression, stability, pathological mutations or post-translational modifications (PS129, N-terminal acetylation) influence neuronal death. Additionally, I will show an assay that we have set up to score for cell-autonomous and non-cell-autonomous alpha-synuclein toxicity.
